Giemsa Stain is a laboratory reagent and stain for microscopy. The stain functions by rearranging chromatin and inducing G bands producing a high quality stain for chromatin and the nuclear membrane. Giemsa Stains have been used for the histopathologic detection of malaria and other parasites such as spirochete and protozoan. The stain has also been incorporated in differential staining between bacterial and human cells. The compound has been used in combination with May-Grunwald or Wright Stain (sc-203763) as a histological hematology stain. However, the May-Grunwald-Giemsa procedure produces a more intense coloration than Wright-Giemsa.

TECHNICAL INFORMATION
Usage : | Technique used to uniformly stain chromosomes that will leave centromeres constricted:
Method 1 materials: 4 mL Giemsa; 96 mL pH 6.8 phosphate buffer procedure: 1.Place slides in a Coplin jar or staining dish. 2.Prepare the working stain and pour it over the slides. Stain for 7 minutes. 3.Rinse slides in two changes of distilled water. 4.Air dry slides; mount them with a cover slip if desired. (If sequential banding procedures are to follow, cover slipping is not recommended.) Method 2 materials: 1.5N HCl 2.Distilled water 3.pH 6.8 phosphate buffer 4. Giemsa stain: 2 mL stock Giemsa; 4 mL pH 6.8 buffer; 92 mL distilled water procedure: 1.Place slides in a Coplin jar containing 5N HCl for 10 minutes at room temperature. 2.Rinse with tap water for 10 minutes. 3.Stain for in Giemsa for 10 minutes. 4.Rinse, dry, and coverslip, if desired. |
