Applications:
Histological Staining: Pyronin Y is widely used in histology to stain nucleic acids, particularly RNA, in tissue samples. It selectively binds to RNA molecules and stains them with a red or pink color. This staining technique allows for the visualization and analysis of RNA distribution and abundance within cells and tissues. Pyronin Y staining can be used alone or in combination with other dyes to provide contrast and aid in the identification of specific tissue structures.Microscopic Imaging: Pyronin Y is compatible with various microscopic imaging techniques, including brightfield and fluorescence microscopy. It is often used to enhance the visualization of cellular components, such as nuclei and cytoplasm, by selectively staining RNA-rich regions. The red or pink color of Pyronin Y allows for easy identification and analysis of RNA-containing structures under the microscope.Biological Staining: Pyronin Y can be used as a vital stain in biological research. It is often employed to label and track RNA within cells and tissues, providing insights into RNA localization, transport, and turnover. It can also be used to stain specific cell types or cellular structures in biological samples.Flow Cytometry: Pyronin Y staining can be used in flow cytometry, a technique used to analyze and sort cells based on their physical and chemical characteristics. It enables researchers to study RNA content or RNA-to-DNA ratios in individual cells within a population. Pyronin Y fluorescence can be detected and quantified using appropriate flow cytometry instrumentation.Other Applications: Pyronin Y has been used in various other applications, including fluorescence-based assays and techniques, RNA quantification assays, and microbiological studies.

Pyronin Y is a dye that intercalates and stains RNA red, and has been shown to accumulate in the mitochondria of viable cells. Multidrug-resistant (MDR) Acinetobacter baumannii studies tested inhibition of the efflux pump by measuring the intracellluar accumulation of Pyronin Y and Ethidium bromide (sc-203947). Research shows that at low Pyronin Y concentrations the cytostatic effect is cell arrest in the G1 phase, whereas at high concentrations the cells arrest is in the G2 and S phase. Pyronin Y has been used in combination with Alcian Blue (sc-214517) to asses and study pulmonary diseases, and in combination with Methyl Green (sc-215348) to stain nucleic acids in paraffin sections. Additionally Pyronin Y has been used to measure RNA in flow cytometry.
