Embedding involves placing a sample in a supportive material, like paraffin or plastic, for cutting thin sections for microscopy. Sectioning is done using specialized blades called microtomes or ultramicrotomes. These sections are then placed on microscope slides and stained. Different staining protocols are available to enhance the visibility of specific structures. The most commonly used stain is a combination of hematoxylin and eosin, known as H&E stain. Hematoxylin stains nuclei blue, while eosin stains cytoplasm pink. Therefore, H&E slides typically show shades of pink and blue. Toluidine blue stains the nucleus and cytoplasm blue, but mast cells appear purple. Wright’s stain colors red blood cells blue/purple, while white blood cells and platelets have different colors. H&E stain provides a permanent stain, allowing slides made with this combination to be preserved for later examination. However, some other histology stains are temporary, so photomicrography is used to preserve the data. Most trichrome stains are differential stains, meaning a single mixture produces multiple colors. For example, Malloy’s trichrome stain colors cytoplasm pale red, the nucleus and muscle red, red blood cells and keratin orange, cartilage blue, and bone deep blue.
